Twitter has drastically reduced the reach of tweets with external links, just like any other social platform. That's why we're seeing so many Twitter threads these days. So if you want to share links directly, do NOT put them into the first tweet but write a meaningful thread.
Even better, though, is to use Twitter the way it is intended to be used - for conversations on the platform. Write tweets that inspire Twitter users to check out your profile and treat your profile like a landing page. You can place a link to your podcast in your bio or in the URL field on your profile.

My best tip is to set up a private list of those accounts that could serve as potential guests or sponsors for your podcast - and engage with these accounts every single week.
By adding them to a private list and following these accounts, you create a separate timeline only for their tweets. It's important that you turn on notifications for new tweets from those accounts so that you're one of the first to respond. That will allow all future respondees to see your tweet in the list of responses and therefore gives more exposure to your response and profile.
Set aside 20-30 minutes at least twice weekly and communicate with those accounts. Build relationships, and you'll see that those accounts will start helping you to promote your podcast on Twitter.
Engage with other tweets.
When you see relevant tweets to your podcast, make sure to like and share them! This will help you get on the radar of those who wrote those tweets, and they may start following you back. And don't forget to @mention people when appropriate - this is a great way to start a conversation.
